Piperazine hexahydrate, also known as 1,4-diazacyclohexane, is a colorless crystal with an ammonia-like odor and strong hygroscopicity. It is soluble in water, methanol, and ethanol, and slightly soluble in benzene.

Piperazine hexahydrate is produced from chloroethanol through amination and cyclization. The process involves heating chloroethanol and ammonium hydroxide in an autoclave until the ammonia pressure reaches 441 kPa, stirring for 5 hours, and recovering excess ammonium hydroxide at atmospheric pressure to obtain an aminoethanol aqueous solution. A measured amount of hydrochloric acid is added with stirring, and the mixture is concentrated under reduced pressure to a semi-paste-like ethanolamine hydrochloride concentrate. The temperature is raised to remove water until 150°C, pre-melted paraffin is added, and the temperature is further increased to 260°C, maintaining a reaction at (260±2)°C for 12 hours. The mixture is discharged and filtered to remove paraffin, yielding piperazine hydrochloride. Piperazine hydrochloride, solid sodium hydroxide, and water are added to a reaction kettle, stirred for 1.5 hours, and ammonia is expelled. The temperature is then raised to 130-140°C to distill off the piperazine hexahydrate solution. The residue is filtered out, and the piperazine hexahydrate solution is cooled to 8°C and centrifuged to obtain piperazine hexahydrate.

Preparation Method:

Using chloroethanol as the raw material, it is produced through amination and cyclization. Uses: Piperazine hexahydrate is soluble in ethanol. Adding glacial acetic acid with stirring and cooling precipitates piperazine acetate, which is used in the synthesis of hormonal drugs such as hydrocortisone phosphate sodium. Piperazine hexahydrate reacts with acetic anhydride to produce acetylpiperazine, an intermediate for the anthelmintic drug piperazine nitrothiazole. Dehydration of piperazine hexahydrate yields anhydrous piperazine.
